Although mention must go to the Redmond Derby, which though not a "major" bike race, has run continuously since 1939, making it the oldest bike race in America. For much of its history it was not ABLA sanctioned, and was probably won at least a few times by a kid on a paperboy bike with fenders, so I don't mind if people call Somerville the oldest "real" bike race in America. http://www.ci.redmond.wa.us/recreationarts/derbydays/criterium.asp
